Biography: Dani Dumitriu, MD, PhD, is trained as a general pediatrician, neuroscientist, and pediatric environmental health scientist. She joined Columbia University as an Associate Professor of Pediatrics (in Psychiatry) in November 2018. She dedicates 80% of her time to basic science research into the neurobiological basis of resilience as the Principal Investigator of the DOOR lab (Developmental Origins of Resilience lab) at the New York State Psychiatric Institute and 20% time to caring for newborns in the Well-Baby Nursery at the Morgan Stanley Children's Hospital of New York. The overarching goal of Dr. Dumitriu's research program is to understand and harness the neurocircuits of resilience to spearhead novel approaches to primary prevention. This is motivated by the observation that during the last century, medicine has made unprecedented progress in the treatment of disease, with a lack of parallel advancement in the prevention of disease. Consequently, the largest current worldwide disease burden comes from preventable mental and physical illnesses, such as depression, anxiety, diabetes, and hypertension. Stressful life events, particularly during critical developmental periods, have been linked to increased risk of virtually all diseases, including metabolic and cardiovascular ones, and a significant effort is underway to understand stress-induced biological consequences. However, even in the presence of the most severe stressors, some individuals remain resistant to the development of stress-induced pathology. Identifying the neurobiological basis for this natural resilience can therefore lead to the development of novel preventative strategies aimed at strengthening resiliency circuits. To tackle the mechanisms for individual variability in stress-responses, the DOOR lab uses several animal models, with the overarching unifying goal of understanding the structure, function, and Developmental Origins of Resilience. In a mouse model of adult social defeat stress, we dissect preexisting neurocircuit structure-function differences in susceptible versus resilient animals using state-of-the-art tools including in vivo chemogenetics, optogenetics, and miniscope imaging, ex vivo super-resolution imaging of dendritic spines, ex vivo whole-brain imaging of functional or structural connectivity using a variety of genetic and/or viral labeling tools, and graph theoretical analysis of co-activation patterns as a proxy for functional connectomics. In a separate set of experiments, we combine rat early-life maternal separation with wireless EEG and/or EKG recordings in dams and their pups. This data is used to understand individual variability in developmental trajectories and the role of dam-pup autonomic synchrony as a potential protective factor. Parallel EKG recordings and synchrony analyses are being conducted during mother-infant interactions in the newborn nursery. Finally, the lab is spearheading a study on wild rat stress-responses, asking the simple question: do depressive-like states occur in the wild, or are they a genetic and environmental epiphenomenon of laboratory breeding? In addition to answering questions about the neurobiological basis for resilience, the DOOR lab is committed to the development of new tools and to the movement toward #openscience. In collaborative work with two different engineering groups, we are developing novel wireless telemetry devices for wild rats and a robotic solution for ex vivo neuronal microinjections. Biography: Dr. Justine Kahn is a Pediatric Oncologist and a clinical investigator specializing in survival outcomes, cancer care delivery, and health equity in children and adolescent/young adults (AYA) with Hodgkin lymphoma, non-Hodgkin lymphoma, acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL). Dr. Kahn received her MD from the Icahn School of Medicine at Mount Sinai in New York and completed her Pediatric Residency at Mount Sinai Kravis Children's Hospital in New York. Dr. Kahn completed her Clinical Fellowship in Pediatric Hematology/Oncology at Columbia University Medical Center. While in residency, Dr. Kahn was elected to Alpha Omega Alpha, the national medical honor society which recognizes high educational achievement. During her fellowship, Dr. Kahn served as a Postdoctoral Research Fellow in Cancer-Related Population Sciences supported by the National Cancer Institute for her work in cancer care delivery, health equity, and health outcomes research. Upon graduation from fellowship, Dr. Kahn joined the Pediatric Hematology/Oncology Division at Columbia University Medical Center as an Oncology Instructor and Attending Physician in 2016. She is currently involved in patient care, clinical trials research, and health outcomes research. Dr. Kahn is actively conducting research through studies developed in the Dana-Farber Cancer Institute (DFCI) ALL Consortium, the Children's Oncology Group (COG), and at Columbia University. Dr. Kahn has chosen to focus her research efforts on underserved populations because studies suggest that poor access-to-care, low health literacy, and financial stress can all negatively impact survival in children and AYAs with cancer. Biography: Monica Bhatia, MD specializes in the care of children with noncancerous blood disorders who may benefit from bone marrow transplantation, especially sickle cell anemia. She and her colleagues are working to reduce the toxicity (side effects) and complications of this treatment without compromising its effectiveness, and to make it available to more patients. The field of pediatric hematology brings with it many intellectual challenges, and bone marrow transplantation in particular is especially dynamic. Dr. Bhatia has the opportunity to provide care for the sickest children, potentially curing their disease. She builds strong relationships with her patients and their families that last for years. It is inspiring for her to see young patients grow up to lead healthy, productive lives. NewYork-Presbyterian/Morgan Stanley Children's Hospital has a robust program for bone marrow transplantation for sickle cell anemia - the only cure for this disease. Dr. Bhatia and her team have witnessed remarkable results among children who are cured with bone marrow transplantation and spared from the potentially life-threatening complications of sickle cell disease. Yet despite the advances made, only 15 percent of patients with sickle cell disease have a matched sibling bone marrow donor. Therefore, Dr. Bhatia and her team conduct research to evaluate a procedure called "CD34-selected stem cell transplantation" to remove T-cells from donor bone marrow for patients without a matched sibling donor. This approach reduces the risk of graft-versus-host disease (GVHD), a potential complication of bone marrow transplantation in which T-cells from the donor attack the tissue of the recipient. This approach to harnessing stem cells could potentially make bone marrow transplantation a safer, effective option for many patients who are seeking a cure for sickle cell disease. Dr. Bhatia and her colleagues are currently assessing reduced-intensity transplantation, which uses lower doses of chemotherapy to decrease the risk of side effects and long-term complications without lowering the effectiveness of treatment. She is also interested in looking for ways to further improve the quality of life of patients who have had a bone marrow transplant in addition to fertility preservation.

This new functional, vascularized lung graft then enabled the attachment and engraftment of lung epithelial progenitors. Dr. Dorrello's laboratory is focused on elucidating the optimal strategies to selectively remove defective lung epithelial cells and supportthe proliferation and differentiation of lung progenitors towards healthy epithelium and ultimately lung repair.
